Modification of cytosine plays an important role in epigenetic regulation of gene expression and genome stability. Cytosine is converted to 5-methylcytosine (5mC) by DNA methyltransferase; in turn, 5mC may be oxidized to 5-hydroxymethylcytosine (5hmC) by ten-eleven translocation enzyme. The structural flexibility of DNA is known to affect the binding of proteins to methylated DNA. Here, we have carried out a semi-quantitative analysis of the dynamics of double-stranded DNA (dsDNA) containing various epigenetic modifications by combining data from imino 1H exchange and imino 1H R1ρ relaxation dispersion NMR experiments in a complementary way. Using this approach, we characterized the base-opening (kopen) and base-closing (kclose) rates, facilitating a comparison of the base-opening and -closing process of dsDNA containing cytosine in different states of epigenetic modification. A particularly striking result is the increase in the kopen rate of hemi-methylated dsDNA 5mC/C relative to unmodified or fully methylated dsDNA, indicating that the Watson-Crick base pairs undergo selective destabilization in 5mC/C. Collectively, our findings imply that the epigenetic modulation of cytosine dynamics in dsDNA mediates destabilization of the GC Watson-Crick base pair to allow base-flipping in living cells.
The objective was to evaluate the cost-effectiveness of one-time opportunistic screening for AF in general practice in citizens aged ≥65 years in Denmark compared to a no-screening alternative following current Danish practice.
A decision tree and a Markov model were designed to simulate costs and quality-adjusted life years (QALYs) in a hypothetical cohort of citizens aged ≥65 years equivalent to the Danish population (1 M citizens) over the course of 19 years, using a healthcare and societal perspective. Share of detected AF patients following opportunistic screening was retrieved from a recent Danish screening study, whereas the risk stroke and bleedings in AF patients were based on population data from national registries and their associated costs was obtained from published national registry studies.The present study showed that one-time opportunistic screening for AF was more costly, but also more effective compared to a no-screening alternative. The analysis predicts that one-time opportunistic screening of all Danes aged ≥65 years potentially can identify an additional 10,300 AF patients and prevent 856 strokes in the period considered. The incremental cost of such a screening program is €56.4 M, with a total gain of 6,000 QALYs, resulting in an incremental cost-effectiveness ratio of €9,400 per QALY gained.

It is therefore important to establish a robust and rapid method of synthesizing polyserotonin nanoparticles (PSeNP) in the size range ideal for in vivo utilization. Since the formation of PSeNP is base-catalyzed, here we report the influence of solution pH, in the presence of different base systems, on the kinetics of PSeNP formation and physico-chemical properties of the resulting nanoparticles. We show that the rate of formation and the size of PSeNP depend on both the nature of the base and the initial pH of the reaction. We have also improved the kinetics of particle formation by performing the synthesis at an elevated temperature (60 °C), leading to a dramatic reduction in synthesis time from days to hours. This presents a significant advance in the efficiency of PSeNP synthesis and provides a facile approach in tuning the size of nanoparticles to suit various applications. Furthermore, we show that similar to serotonin, PSeNP also exhibits free radical scavenging property. Our results demonstrate that PSeNP has the potential to become a key player in the advancement of nanotechnology-mediated antioxidative therapy.A simple yet effective soaking treatment has been proposed to fabricate hydrogels with desirable mechanical properties, but the strengthening mechanism of hydrogels lacks an in-depth study. Here, we investigated the influence of kosmotropic citrate anion on the structure and properties of immersed gelatin hydrogels. The obtained hydrogels possessed the properties of high strength, modulus and toughness simultaneously. The dehydration of hydrogels facilitated the interactions among gelatin molecules, resulting in the formation of helix structures. Both the content and length of the triple helices increase with an increase in citrate concentration, which in turn contributes to the strengthening of hydrogels. The excellent mechanical performances of these hydrogels may open up new applications for protein materials.The application of functional self-healing and mechanically robust hydrogels in bioengineering, drug delivery, soft robotics, etc., is continuously growing. However, fabricating hydrogels that simultaneously possess good mechanical and self-healing properties remains a challenge. Developing robust hydrogel formulations for the encapsulation and release of hydrophobic substances is a major challenge especially in some pharmaceutical treatments where the many of drugs show incompatibility with the hydrophilic hydrogel matrices. Schiff base hydrogels have been developed using a benzaldehyde multifunctional amphiphilic polyacrylamide crosslinker in conjunction with glycol chitosan. The polymeric crosslinker was synthesized by a two-step reaction using aqueous Cu-RDRP to give an ABA telechelic copolymer of N,N-dimethyl acrylamide (DMAc) and N-hydroxyethyl acrylamide (HEAm) from a bifunctional PEG. The polymer was then modified by post functionalization leading to a multifunctional benzaldehyde crosslinker that was shown to be capable of self-assembly into aggregates in aqueous media serving as a possible candidate for the entrapment of hydrophobic substances.
