Treatment of cells with antioxidant molecules leads to a dose-dependent decrease of intracellular H2O2 levels. As a proof of concept, the bioassay has been used to measure the antioxidant activity of extracts from Brassica juncea wastes, which contain glucosinolates, isothiocyanates and other antioxidant molecules. In as short as 3 months, COVID-19 has spread and ravaged the world in an unprecedented speed in modern history rivaling the 1918 flu pandemic. SARS-CoV-2, the culprit virus, is highly contagious and stable in the environment and predominantly transmits among humans via the respiratory route. Accumulating evidence suggest that this virus, like many of its related viruses, may also be an enteric virus that can spread via the fecal-oral route. Such a hypothesis would also contribute to the rapidity and proliferation of this pandemic. Here we briefly summarize what is known about this family of viruses and literature basis of the hypothesis that SARS-CoV-2 is capable of infecting the gastrointestinal tract and shedding in the environment for potential human-to-human transmission. PURPOSE To report the outcomes of initial high-dose and extended taper of antiviral and steroid prophylaxis for the treatment of eyes with high-risk vascularized herpetic corneal scars that underwent two-piece mushroom keratoplasty (MK). DESIGN Prospective interventional case series. METHODS In this single-center study, 52 consecutive eyes with vascularized (≥2 quadrants) herpetic corneal scars underwent 2-piece microkeratome-assisted mushroom keratoplasty. Initial high-dose and extended taper of combined oral and topical antiviral and steroid prophylaxis was administered. Outcome measures were best spectacle-corrected visual acuity (BSCVA), refractive astigmatism (RA), endothelial cell density, immunologic rejection, herpetic recurrence and graft failure rates. RESULTS Excluding patients with vision-impairing comorbidities, baseline BSCVA (1.73±0.67 logMAR) significantly improved annually during the first 2 years (p less then 0.001, p = 0.016), reaching 0.17±0.18 logMAR at year 2, and remaining stable up to 10 years (p = 0.662). At 2 years, 86% of eyes saw ≥20/40, 55% saw ≥20/25 and 18% saw ≥20/20 Snellen BSCVA. RA exceeded 4.5 diopters in 7% of cases after wound revision for high-degree astigmatism in 7 cases. Endothelial cell loss was 40.9% at 1 year with an annual decline of 3.1% over 10 years. TEPP-46 cell line The 10-year cumulative risk for immunologic rejection, herpetic recurrence and graft failure was 9.7%, 7.8% and 7.6%, respectively. CONCLUSION Initial high-dose and extended taper of antiviral and steroid prophylaxis in mushroom keratoplasty for high-risk, vascularized herpetic corneal scars achieves clinical outcomes that remain stable for up to 10 years after surgery with minimal risk of immunologic rejection, herpetic recurrence and graft failure. Depression is a mental illness with a high prevalence in humans reaching 21% of the worldwide population.The present study aims to evaluate the antidepressant effect of different formulations of Thymoquinone; free Thymoquinone (TQ), Thymoquinone-loaded Chitosan nanoparticles (TQ-TPP-Cs NPs) and Thymoquinone-loaded Chitosan nanoparticles coated with polysorbate 80 (TQ-TPP-Cs NPs-PSb80) that have been prepared to avoid the low bioavailability of TQ. Rats were randomly separated into control rats, depression control induced by reserpine, rat model treated with TQ, rat model treated with TQ-TPP-Cs NPs and rat model treated with TQ-TPP-Cs NPs-PSb80. The results indicate that TQ-TPP-Cs NPs loaded with polysorbate 80 was more efficient in ameliorating the behavioral and neurochemical changes induced by reserpine than TQ and TQ-TPP-Cs NPs. Formulationswere characterized for size, morphology, encapsulation efficiency and in vitro drug release before their use in treatment. Reserpine induced a reduction in motor activity and swimming time and increased immobility time as indicated from the open field test (OFT) and forced swimming test (FST). In addition, a significant decrease in the monoamine neurotransmitters serotonin (5-HT), norepinephrine (NE) and dopamine (DA) was recorded in the cortex, hippocampus and striatum of reserpine-treated rats. The present data suggest that the antidepressant efficacy of TQ could be enhanced by engaging TQ with chitosan nanoparticles as a drug carrier and the formulations were modified by coating with polysorbate 80. The crayfish plague pathogen Aphanomyces astaci, which is among the most studied pathogens of aquatic invertebrates, co-evolved with North American crayfish species but threatens crayfish on other continents. The pathogen causes mass mortalities, particularly in Europe. In this study we document 12 crayfish plague outbreaks that occurred from 2014 to 2019 in Czechia and, by using available molecular techniques (microsatellite and mtDNA markers), we reveal the A. astaci genotypes involved. Our results provide the first evidence of strains from genotype group D, originally associated with the host Procambarus clarkii, causing Astacus astacus and Austropotamobius torrentium mass mortalities in Czechia. Moreover, mtDNA sequencing confirmed two distinct haplotypes of the D haplogroup, indicating two independent sources of infection, presumably originating from ornamental crayfish in the pet trade or spreading from crayfish established in neighbouring countries. Genotype group A was recorded in two A. astacus mortalities, and genotype group E, associated with Faxonius limosus, in two A. torrentium and three A. astacus mortalities. Microsatellite genotyping also reidentified the unusual genotype SSR-Up in two A. astacus outbreaks, ten years after its first documented occurrence. In addition, we tested healthy-appearing indigenous crayfish from 25 localities for potential chronic infections. No traces of A. astaci DNA were detected; chronic infections in European crayfish species thus do not seem a pervasive phenomenon in Czechia. However, their role as A. astaci latent reservoirs, especially in Pontastacus leptodactylus populations introduced to the country since the late 19th Century, cannot be excluded.
