The favorable thermodynamic and kinetic conditions are the powerful force to drive the rapid lift upwards and infusion of molten Li into CNTs network, which is the key to form a uniform metallic layer in Li/CNTs hybrid. The obtained hybrid indicates super-stable functions even at an ultrahigh current density of 40 mA cm-2 for 2000 cycles with a stripping/plating capacity of 2 mAh cm-2 in symmetric cells. Subsequently, this hybrid also demonstrates a significantly decreased resistance, excellent cycling stability at high current density and flexibility in the full Li-S battery. Many natural proteins have undergone directed evolution in vitro in the test tubes in the laboratories worldwide, resulting in the numerous protein variants with novel or enhanced functions. we constructed here an SH2 variant library by randomizing 8 variable residues in its phosphotyrosine (pTyr) binding pocket. Selection of this library by a pTyr peptide led to the identification of SH2 variants with enhanced affinities measured by EC50. Fluorescent polarization was then applied to quantify the binding affinities of the newly identified SH2 variants. As a result, three SH2 variants, named V3, V13 and V24, have comparable binding affinities with the previously identified SH2 triple-mutant superbinder. Biolayer Interferometry assay was employed to disclose the kinetics of the binding of these SH2 superbinders to the phosphotyrosine peptide. The results indicated that all the SH2 superbinders have two-orders increase of the dissociation rate when binding the pTyr peptide while there was no significant change in their associate rates. To identify and assess demographic, sonographic and Proactive Molecular Risk Classifier for Endometrial cancer (ProMisE) prognostic factors for recurrence or progression in endometrial cancer (EC).
We prospectively included 339 women with EC, undergoing expert transvaginal ultrasound before surgery. Tumors were classified according to FIGO, and ProMisE (MMR-D, POLE EDM, p53wt and p53abn). ProMisE subtypes were compared regarding demographic, sonographic characteristics, recurrence or progression, and survival. Cox regression was used to identify prognostic factors associated with recurrence or progression, with univariable models to study crude associations and multivariable models to study adjusted associations. Logistic regression and ROC curves analysis was used to assess the predictive ability of the prognostic factors, regarding recurrence or progression within three years, and to compared their predictive ability to that of the European Society for Medical Oncology (ESMO) classification. In separateic and ProMisE prognostic factors had higher ability to predict recurrence or progression than the ESMO classification, supporting their use in preoperative risk stratification. The p53 status combined with ultrasound tumor size has the potential to preoperatively identify a large group of women with a very low risk of recurrence or progression. This article is protected by copyright. All rights reserved.Laryngeal squamous cell carcinoma (LSCC) is the most common malignant tumor, which occurs in the head and neck. Current treatments for LSCC are all largely weakened by increasing drug resistance. Our study aimed to investigate the effects of long noncoding RNA (lncRNA) H19 on drug resistance in LSCC. In our study, we found that the level of H19 was sharply upregulated in LSCC tissues and drug-resistant cells compared with the control. Besides, the expression of high-mobility group B1 (HMGB1) was elevated, and microRNA107 (miR-107) was suppressed in drug-resistant cells compared with the control. Further study revealed that the interference of H19 by short hairpin RNA (shRNA) effectively suppressed high autophagy level and obvious drug resistance in drug-resistant cells. Besides that, miR-107 was predicted as a target of H19 and inhibiting effects of H19 shRNA on autophagy and drug resistance were both reversed by miR-107 inhibitor. Moreover, HMGB1 was predicted as a target of miR-107 in LSCC cells and knockdown of HMGB1 was able to suppress autophagy and drug resistance in LSCC cells. In addition, our investigation demonstrated that H19 shRNA exerted an inhibiting effect on autophagy and drug resistance by downregulating HMGB1 by targeting miR-107. Finally, the in vivo experiment revealed that LV-H19 shRNA strongly suppressed drug resistance compared with the usage of cisplatin individually. Taken together, our research indicated an H19-miR-107-HMGB1 axis in regulating the autophagy-induced drug resistance in LSCC in vitro and in vivo, providing novel targets for molecular-targeted therapy and broadening the research for LSCC.
