We showed that AMP-activated protein kinase (AMPK) and endosomal-sorting-complex-required-for-transport (ESCRT) factors are required for proteasome microautophagy and also impact PSG dissipation and nuclear reimport of proteasomes after glucose refeeding. The insoluble protein deposit (IPOD) compartment provides an alternative means of proteasome homeostasis, including when microautophagy is impaired. Our findings reveal a surprising diversity of mechanisms for proteasome quality and quantity control during starvation. A mechanistic understanding of the AMPK-regulated ESCRT-mediated microautophagy pathway could provide new avenues for manipulating proteasome homeostasis and treating human disease.OBJECTIVES To describe our experience with men admitted to a tertiary care hospital with genital injury. METHODS Adult men with injuries of the genitals, admitted to our institution between January 2013 and June 2018, were identified from our institutional trauma registry. Patient charts were queried to extract mechanism, management, follow-up, and complications. RESULTS 118 men met inclusion criteria. 39% and 61% sustained penetrating and blunt injuries, respectively. The most common mechanisms of penetrating trauma were external violence (48%) and self-inflicted injury (40%). The most common mechanisms of blunt trauma were motorcycle crash (33%) and sexual injury/intercourse (22%). 38% presented with penile and 71% with scrotal injuries. 48% of men with scrotal injuries had concomitant testis injury. 9.3% presented with both a penile and a scrotal injury. Concomitant urethral injuries were found in 17% of all genital injuries. Genital trauma was more common in the summer months. 74% of all genital injuries were managed operatively, with surgery more common after penetrating injury (89% vs 64%, p value  less then  0.01). Epacadostat 73% of 84 men with scrotal trauma were managed operatively. 27 men received surgical intervention for testis rupture, with a testicular salvage rate of 44%. 60 (51%) patients presented for follow-up. The median length of follow-up from initial injury was 29 (± 250) days. Of these, 9 (15%) patients developed one or more complications CONCLUSIONS Genital injuries can occur via numerous mechanisms and frequently require operative intervention. Concomitant urethral injury is common. More work is needed to evaluate the long-term sequelae of these injuries.Whisker touch is an active sensory system. Previous studies in Pinnipeds have adopted relatively stationary tasks to judge tactile sensitivity, which may not accurately promote natural whisker movements and behaviours. This study developed a novel feeding task, termed fish sweeping to encourage whisker movements. Head and whisker movements were tracked from video footage in Harbor seal (Phoca vitulina), California sea lion (Zalophus californianus) and Pacific walrus (Odobenus rosmarus divergens). All species oriented their head towards the moving fish target and moved their whiskers during the task. Some species also engaged in whisker control behaviours, including head-turning asymmetry in the Pacific walrus, and contact-induced asymmetry in the Pacific walrus and California sea lion behaviours that have only previously been observed in terrestrial mammals. This study confirms that Pinnipeds should be thought of as whisker specialists, and that whisker control (movement and positioning) is an important aspect of touch sensing in these animals, especially in sea lions and walruses. That the California sea lion controls whisker movement in relation to an object, and also had large values of whisker amplitude, spread and asymmetry, suggests that California sea lions are a promising model with which to further explore active touch sensing.Selenium nanoparticles (Se NPs) were synthesized using Saccharomyces cerevisiae yeast. Influences of different amounts of sodium selenite (5.0, 10.0, 15.0, 20.0, and 25 µg) were evaluated on growth of yeast during incubation at 32 °C, during 4 days. UV-Vis spectroscopy results have shown that synthesized Se NPs had broad emission peak (λmax) in the wavelength around 350 nm which demonstrated that formation of Se NPs occurred in intracellular manner. Physico-chemical characteristics of the synthesized Se NPs using dynamic light scattering particle-size analyzer indicated that the fabricated Se NPs had particle size, polydispersity index, and zeta potential ranging from 75 to 709 nm, 0.189 to 0989, and -7.06 to -10.3 mV, respectively. Obtained results revealed that intracellular Se NPs with minimum particle size (75 nm), maximum zeta potential (-10.3 mV), and antioxidant activity (48.5%) were synthesized using minimum amount of selenium salt (5 µg). However, most uniform Se NPs were formed using maximum amount of selenium salt (25 µg). Results also indicated that by increasing amount of sodium selenite in the culture media, from 5.0 to 25 µg, antioxidant activity of the formed Se NPs decreased from 48.5 to 20.8, respectively.Permanent reclamation of tailings generated by surface mining in the Canadian oil sands may be achieved through the creation of end pit lakes (EPLs) in which tailings are stored in mined-out pits and capped with water. However, these tailings contain high concentrations of dissolved organics, metals, and salts, and thus surface water quality of EPLs is a significant concern. This is the first study to investigate the chronic toxicity of surface water from Base Mine Lake (BML), the Canadian oil sands first large-scale EPL, to aquatic invertebrates that play a vital role in the early development of aquatic ecosystems (Chironomus dilutus and Ceriodaphnia dubia). After exposure of C. dilutus larvae for 23 days and C. dubia neonates for 8 days, no mortality was observed in any treatment with whole BML surface water. However, the emergence of C. dilutus adults was delayed by nearly 1 week, and their survival was significantly reduced (36%) compared with the controls. Reproduction (fecundity) of C. dubia was reduced by 20% after exposure to 2014 BML surface water; however, the effect was not observed after exposure to BML surface water collected a year later in 2015. Despite some adverse effects, the results of this study indicate that BML surface water quality is improving over time and is able to support certain salt-tolerant aquatic organisms. Because salinity within BML will persist for decades without manual intervention, the ecological development of the lake will likely resemble that of a brackish or estuarine ecosystem with reduced diversity.
