Amyotrophic Lateral Sclerosis is a progressive, incurable amyloid aggregating neurodegenerative disease involving the motor neurons. Identifying potential biomarkers and therapeutic targets can assist in the better management of the disease. We used an integrative approach encompassing analysis of transcriptomic datasets of human and mice from the GEO database. Our analysis of ALS patient datasets showed deregulation in Non-alcoholic fatty acid liver disease and oxidative phosphorylation. Transgenic mice datasets of SOD1, FUS and TDP-43 showed deregulation in oxidative phosphorylation and ribosome-associated pathways. Commonality analysis between the human and mice datasets showed oxidative phosphorylation as a major deregulated pathway. Further, protein-protein and protein-drug interaction network analysis of mitochondrial electron transport chain showed enrichment of proteins and inhibitors of mitochondrial Complex III and IV. The results were further validated using the yeast model system. Inhibitor studies using metformin (Complex-I inhibitor) and malonate (Complex-II inhibitor) did not show any effect in mitigating the amyloids, while antimycin (Complex-III inhibitor) and azide (Complex-IV inhibitor) reduced amyloidogenesis. Knock-out of QCR8 (Complex-III) or COX8 (Complex-IV) cleared the amyloids. Taken together, our results show a critical role for mitochondrial oxidative phosphorylation in amyloidogenesis and as a potential therapeutic target in ALS.Communicated by Ramaswamy H. Sarma.Objectives This study aims to determine the sociodemographic characteristics of pet owners and their sources of information regarding pet-related health risks. It also assesses pet owners' perception of pet-related health risks and evaluates their attitudes and practices regarding preventative measures toward pet-related health risks. Methods A cross-sectional study was conducted in Riyadh, Saudi Arabia from September 2020 to February 2021. The target population was pet owners visiting pet stores. The city was divided into five equal sectors and data were collected from each sector using a questionnaire. Results A total of 508 participants were engaged in the study. Of these, 402 participants fit the inclusion criteria. The mean age of the participants was 25.5 years. Gender was almost equally distributed, and the majority of participants were single, however most of the participants lived with four or more family members. Around 49.5% of participants had household members at a higher risk for pet-related diseases. The most common pet types were cats (77.4%), followed by birds (24.6%) and dogs (9.7%). Most participants were satisfied with their understanding of pet-related health risks and were not concerned about getting sick because of contact with pets. Physical harm was the most common pet-related health risk that the participants were satisfied with their level of understanding about, on the other hand their satisfaction with their understanding about gastrointestinal infections from pet contact was the lowest. Most participants had good pet husbandry and hygiene practices. Social media and veterinarians were the most common sources of information regarding pet-related health risks. Conclusion Although most pet owners reported having good pet husbandry practices, further education is needed to raise awareness about pet-related health risks. Since most pet owners receive their information about this topic from social media and veterinarians, further education through these platforms might be beneficial.
The burden of the COVID-19 pandemic is often enumerated in lives lost, but the strain on health care resources and mobility limitations contributed to the burden of non-COVID related disease. In this study, we evaluated the impact of the pandemic through a time series review of cytology samples.
Pathology reports for all cytology specimens received from January 2019 through April 2021 at our institution were reviewed. Time series analysis was performed using moving averages, time trend analysis, cross-correlation, and tests of homogeneity.
During the first peak of the pandemic (March-June 2020), breakpoint analysis showed a downward shift in the number of gynecologic (-89.4%) and non-gynecologic (-70.4%) cytology specimens within a week of declaration of an emergency. Cross-correlation analysis showed a relationship between sample numbers and COVID-19 cases during the initial phase of the pandemic (April-June 2020). Selleck U0126 During the second surge (October 2020-April 2021), despite the higher incidence of COVID-19, there was a smaller impact on cytology samples (-20.1% and - 24.8% for gynecologic and non-gynecologic samples, respectively). Although specimen numbers slowly returned to baseline following the first wave of the pandemic, the earlier decline in malignant diagnoses was not offset during the study period.

There is a great need for construction of a highly reliable and stable electrocatalyst for the efficient detection of cholesterol. In this work, mesoporous NiCo2S4nanoflakes of enhanced electrochemical properties are prepared through a facile hydrothermal approach. The developed nanoflakes modified nickel foam electrode exhibits outstanding electrocatalytic properties for the detection of cholesterol with high selectivity. The electrode displays excellent sensitivity of 8623.6μA mM-1cm-2, in the wide linear range from 0.01 to 0.25 mM with a low detection limit of 0.01μM. In addition, NiCo2S4structure reveals good thermal stability and reproducibility over a period of 8 weeks. Moreover, the nanoflakes show good response for detection of cholesterol in real samples. Our results demonstrate the potential use of NiCo2S4as a catalyst for the development of cost-effective electrochemical sensors for medical and industrial applications.Nitrogen isotopes and abundances in sedimentary rocks have become an important tool for reconstructing biogeochemical cycles in ancient ecosystems. In this study, a systematic review was conducted to collect, meta-analysis and carcinogenic risk assessment of the quantitative data regarding the prevalence and concentration of AFM1 in several types of milk produced in Iran.
In our study, the concentration and prevalence of AFM1 in different types of milk (Raw, pasteurized, and UHT) from 113 original articles in Iran using searching the Web of Science, PubMed, Scopus, Science Direct and Google Scholar (in Persian and English) databases from 2002 to august 2021 were collected. The concentration of AFM1 was meta-analyzed using the random effect model (REM) based on type of milk (raw, pasteurized and UHT) subgroups and Monte Carlo simulation (MCS) approach was used to assess safety risks and investigate carcinogenic nd carcinogenic properties is a public health concern that the results show that the risk of carcinogenesis is higher at younger ages (less than 20 years). As a result, there is a strong association between consumption of raw milk, pasteurized milk and UHT and the risk of cancer in children and adults in Iran.
The purpose of this study was to investigate scotopic contour deformation detection (sCDD), and its structural determinants, in participants with intermediate age-related macular degeneration (iAMD) with or without reticular pseudodrusen (RPD).
Forty-one participants (aged 58-89 years), including 9 with iAMD and RPD, 16 with iAMD only, and 16 controls, underwent functional testing. The sCDD was evaluated with radial frequency arcs presented at 4 loci ±4 degrees and 8 degrees vertical eccentricity. Scotopic thresholds and dark adaptation (DA) were measured at the same loci. Retinal layers of spectral domain optical coherence tomography (SD-OCT) volume scans were segmented. To establish the concurrent validity of the functional test, we evaluated the fraction of variability in sCDD thresholds explained by SD-OCT data.
The iAMD group had significantly worse sCDD thresholds compared with controls (8 degrees inferior retina P = 0.004 and the 4 degrees loci P < 0.02 for both). Elevated sCDD thresholds were risk of AMD progression.
The purpose of this study was to determine whether accommodation-induced changes in ciliary muscle dimensions vary between emmetropes and myopes, and the effect of the image analysis method.
Seventy adults aged 18 to 27 years consisted of 25 people with emmetropia (spherical equivalent refraction [SER] +0.21 ± 0.36 diopters [D]) and 45 people with myopia (-2.84 ± 1.72 D). There were 23 people with low myopia (>-3 D) and 22 people with moderate myopia (-3 to -6 D). Right eye ciliary muscles were imaged (Visante OCT; Carl Zeiss Meditec) at 0 D and 6 D demands. Measures included ciliary muscle length (CML), ciliary muscle curved length (CMLarc), maximum ciliary muscle thickness (CMTmax), CMT1, CMT2, and CMT3 (fixed distances 1-3 mm from the scleral spur), CM25, CM50, and CM75 (proportional distances 25%-75%). Linear mixed model analysis determined effects of refractive groups, race, and demand on dimensions. Significance was set at P < 0.05.
Myopic eyes had greater CML and CMLarc nasally than emmetronger and thicker ciliary muscles than people with emmetropia. During accommodation, the anterior muscle thickened and the curved nasal muscle length shortened, more in myopic than in emmetropic eyes. The fixed distance method is recommended for repeat measures in the same individual. The proportional distance method is recommended for comparisons between refractive groups.
