Adequate characterization of chemical entities made for biological screening in the drug discovery context is critical. Incorrectly characterized structures lead to mistakes in the interpretation of structure-activity relationships and confuse an already multidimensional optimization problem. Mistakes in the later use of these compounds waste money and valuable resources in a discovery process already under cost pressure. Left unidentified, these errors lead to problems in project data packages during quality review. At worst, they put intellectual property and patent integrity at risk. We describe a KNIME workflow for the early and automated identification of these errors during registration of a new chemical entity into the corporate screening catalog. This Automated Structure Verification workflow provides early identification (within 24 hours) of missing or inconsistent analytical data and therefore reduces any mistakes that inevitably get made. Automated identification removes the burden of work from the chemist submitting the compound into the registration system. No additional work is required unless a problem is identified and the submitter alerted. Before implementation, 14% of samples within the existing sample catalog were missing data on initial pass. A year after implementation, only 0.2% were missing data.In this article, using a quantitative approach, we analyze interpersonal, inmate-on-inmate physical violence in Chilean prisons, using administrative records on collective violence, provided by the Prison Service for the period 2014 to 2017. Violence behind bars is problematic as it threatens inmates' fundamental rights such as personal safety but also because it undermines efforts to maintain an environment prone to inmates' social reintegration. Our data showed a sharp increase in the number of collective fights, from 808 in 2014 to more than 4,000 in 2017. In terms of the predictors, being in a private prisons as well as a greater ratio of inmates to guards were associated with increased collective fights for each of the 4 years we examined. Two additional predictors were statistically significant, yet only for 2017 A higher average criminal involvement score and a smaller ratio between inmates/staff were both associated with increased violence. Despite the fact that prison violence has not yet reached the scale or level of brutality that can be seen in other parts of the region (i.e., Brazil), there are signs of concern that authorities should take into account, particularly the rapid increase in collective fights in just a 4-year period and the recent social turmoil that has taken place in Chile, whose impact cannot be seen yet from these data. In terms of recommendations, we suggest that authorities should prioritize efforts in four areas (a) to gather better data on prison violence, including some reliable data on importation variables (age, criminal history, and nationality); (b) to provide prison guards with tools to anticipate and mediate conflicts; (c) to revise and possibly modify the way prisoners are transferred to different facilities, and (d) in sum, to promote prison environments that can be more legitimate and supportive to inmates' reintegration.Objectives Signal transduction through the mothers against decapentaplegic (SMAD) is a family of signal transduction factors that mediate signalling of the transforming growth factor B (TGFB)-superfamily of cell regulatory proteins. A recent transcriptomic analysis of post-mortem, cortical tissue from subjects with schizophrenia found decreased mRNA expression of SMAD2 and SMAD4 in the dorsolateral prefrontal cortex (DLPFC) associated with the disorder. To expand this initial finding, we sought to determine whether SMAD2 and SMAD4 protein were also altered in the cortex from subjects with schizophrenia.Methods Western blotting was used to measure SMAD2 and SMAD4 protein levels in DLPFC and anterior cingulate cortex (ACC) taken post-mortem from subjects with schizophrenia (n = 20) and matched control (n = 20) subjects.Results Compared to controls, levels of SMAD4 were 25% lower in the DLPFC and 38% lower in the ACC from subjects with schizophrenia. By contrast, SMAD2 levels were not altered in either DLPFC or ACC.Conclusions Our finding of lower SMAD4 protein in the cortex suggests there are likely to be abnormalities in cortical TGFB-superfamily signalling in schizophrenia.In Latin America, there is an increasing interest in the implementation and dissemination of evidence-based, family-centered interventions to prevent youth behavioral problems. While families' participation in interventions is integral to achieving the interventions' desired impact, little is known about what predicts Latin American families' attendance. The current study provides a unique opportunity to explore the participation of families living in the United States, Ecuador, and Chile in an evidence-based intervention, Familias Unidas. We tested for differences in attendance rates, family functioning variables, and adolescent behavioral problem variables, then applied a hierarchical multiple regression to (a) identify which variables significantly predicted program attendance and (b) assess whether the country in which the intervention was implemented in moderated the relationship between predictors and program attendance. On average, Chilean and Ecuadorian parents were more engaged and attended more sessions than parents living in the United States. learn more Across samples, there was significant differences in family functioning and adolescent behavioral problem variables. However, effective parent-adolescent communication was the only significant predictor of lower program attendance. A significant interaction effect revealed that even though Chilean parents had high parent-adolescent communication, they were more likely to attend sessions, compared to parents living in the United States. We highlight the promise of engaging and retaining families, across U.S. and Latin American samples, into a culturally syntonic, family-based intervention, and discuss potential explanations for success in Chile and Ecuador. Researchers interested in implementing interventions in Latin America could utilize these findings to better target participants and intervention efforts.
